Overview

GPI-80 is a glycosylphosphatidylinositol (GPI)-anchored protein of approximately 80 kDa, primarily expressed on human neutrophils and to a lesser extent on monocytes, with gene identity VNN2 encoding vanin-2, a pantetheinase family member possessing hydrolase activity. It physically associates with beta2 integrin (CD18/CD11b, Mac-1) on resting neutrophils, regulating beta2 integrin-mediated neutrophil adherence and migration by concentrating on pseudopodia during chemotaxis without sustained integrin association in later migration phases, and sometimes associating with uPAR. GPI-80 is located on plasma membranes and in secretory vesicles; TNF-alpha stimulation induces its release into soluble form in a beta2 integrin adherence-dependent manner involving actin reorganization, tyrosine kinases, p38 MAPK, and intracellular redox changes, observed in rheumatoid arthritis synovial fluid. Antibodies like clone 3H9 inhibit neutrophil adherence, enhance chemotaxis, and distinguish neutrophils from basophils/eosinophils. It also marks primitive cord blood CD34+CD38- and CD34- hematopoietic stem/progenitor cells, and its gene expression correlates with thymoma stage.
