Overview

The gp100:154-162 peptide–HLA-A*0201 complex is a specific combination of a nonamer peptide epitope (IMDQVPFSV, amino acids 154–162 of the gp100/Pmel17 protein) bound to the human MHC class I molecule HLA-A*0201. This complex is selectively presented on the surface of melanoma cells that express both gp100 and HLA-A*0201, as well as on normal melanocytes. Recognition of this complex by cytotoxic T cells can result in targeted cell killing, making it a prominent focus for adoptive T cell therapies and peptide vaccine trials for melanoma patients expressing HLA-A*0201. gp100 and related antigens like MART-1 are frequent targets in immunotherapy due to their high expression in melanoma and ability to generate strong, antigen-specific T cell responses in the context of HLA-A*0201.

Mechanism of action

Induction of antigen-specific cytotoxic T lymphocyte (CTL) response against melanoma cells presenting gp100:154-162 peptide in the context of HLA-A*0201. Targeting by engineered TCRs specific to this peptide–HLA complex induces melanoma cell lysis.
