Overview

The gp100-specific T cell receptor (TCR) is a heterodimeric surface protein found on CD8+ T cells that recognizes the gp100 (PMEL) peptide presented by MHC Class I molecules, specifically HLA-A*02:01 (Source 1.3.1, 1.4.4). gp100 is a melanocyte-differentiation antigen that is highly overexpressed in cutaneous and uveal melanomas, making it a significant target for cancer immunotherapy (Source 1.3.1, 1.3.5). Therapeutic approaches targeting this receptor-ligand interaction include TCR-engineered T cell (TCR-T) therapies and bispecific T cell engagers like tebentafusp (Source 1.3.2, 1.4.1). Tebentafusp consists of a high-affinity soluble gp100-specific TCR fused to an anti-CD3 scFv, which redirects polyclonal T cells to kill gp100-positive tumor cells (Source 1.3.3, 1.3.5). Clinical use of these therapies is restricted to HLA-A*02:01-positive patients and is associated with on-target toxicities such as skin-related adverse events and systemic cytokine release syndrome (Source 1.3.1, 1.3.4). This receptor plays a critical role in the immune system's ability to identify and eliminate malignant melanocytes by sensing intracellularly derived peptides (Source 1.5.3).
