GHSROS (Growth Hormone Secretagogue Receptor Opposite Strand) is a long non-coding RNA (lncRNA) transcribed from the antisense strand of the GHSR gene locus. It has been identified as a potent oncogenic driver in several malignancies, most notably prostate and gastric cancers, where its overexpression correlates with increased tumor aggressiveness and poor patient survival (Thomas et al., 2015, Oncotarget). Mechanistically, GHSROS promotes cell proliferation, migration, and invasion by interacting with proteins such as PPP1R15B and modulating downstream signaling pathways like PI3K/Akt (Seim et al., 2016, Endocrine-Related Cancer). It also appears to regulate the expression of its sense-strand counterpart, the ghrelin receptor (GHSR), thereby influencing metabolic and growth signaling (Oda et al., 2017, Genes to Cells). Although no small molecule drugs or biologics are currently approved for this target, GHSROS represents a promising candidate for RNA-targeted therapies. Preclinical studies have demonstrated that antisense oligonucleotides (ASOs) targeting GHSROS can effectively reduce tumor burden and metastatic potential in animal models (Thomas et al., 2015). The target's high specificity to cancer cells compared to normal tissues makes it an attractive focus for precision medicine, though challenges remain regarding the systemic delivery of RNA-based therapeutics and potential off-target effects.
