Guanine is one of the four main nucleobases in DNA. The O6 position refers to the oxygen atom attached to the sixth carbon of guanine's purine ring. Methylation or other modifications at this position, especially the formation of O6-methylguanine, are strongly linked to mutation induction and cancer initiation due to altered base pairing preferences (pairing with thymine instead of cytosine).
