Overview

Guanine nucleotide-binding protein G(i) subunit alpha-2 (Gαi2), encoded by the GNAI2 gene, is a critical alpha subunit of the heterotrimeric G protein complex (UniProt). It primarily functions as a signal transducer that inhibits adenylate cyclase activity, thereby reducing intracellular cyclic AMP (cAMP) levels (PubMed). Gαi2 is ubiquitously expressed and mediates signaling for a wide array of G protein-coupled receptors (GPCRs), including those for chemokines, neurotransmitters, and hormones (GeneCards). In clinical contexts, Gαi2 is recognized as a proto-oncogene; its overexpression is linked to increased cell proliferation and migration in various malignancies, such as ovarian and gastric cancers (NIH). Beyond oncology, mutations in GNAI2 are associated with cardiovascular conditions like familial ventricular tachycardia and complex immune phenotypes involving both immunodeficiency and autoimmunity (PubMed, Wikipedia). While most current therapies target upstream GPCRs, Gαi2 itself is an emerging target for direct pharmacological intervention in cancer and inflammatory diseases (ResearchGate).
