Guanylate cyclase 2F, retinal (GUCY2F) is a membrane-bound enzyme predominantly localized in retinal photoreceptors, where it catalyzes the production of cyclic GMP (cGMP) needed for phototransduction and recovery of the visual signal after light activation[1][5]. GUCY2F participates in maintaining the dark state in rods and cones by replenishing cGMP after its light-induced hydrolysis, enabling proper visual signaling. Loss-of-function mutations in GUCY2F are associated with X-linked retinitis pigmentosa, cone-rod dystrophy, and Leber congenital amaurosis, making it clinically relevant for inherited blindness syndromes[1][2]. There is no evidence of approved drugs targeting GUCY2F, but its molecular function and disease association render it a putative therapeutic and diagnostic target for ocular diseases as well as being implicated in some cancers[4].
