Overview

The "gut mucosal surface" consists of an organized set of barriers—mucus layers composed predominantly of mucins (mainly MUC2), a monolayer of intestinal epithelial cells with tight junctions, and the underlying lamina propria rich in immune cells. This structure serves to physically separate the internal host environment from the microbial-rich lumen while also participating in dynamic immune sensing, antigen sampling (via M cells), and production of antimicrobial molecules. The "pathogen membranes" are simply the outer lipid or protein envelopes that form the physical barrier of bacteria, viruses, and other microorganisms that interact with or invade the mucosal interface. Dysfunction or breakdown of this barrier, or pathogen strategies to breach it, play key roles in diseases such as inflammatory bowel disease, infection, and certain cancers. This construct is not a precise, singular molecular target but a complex, functional interface that is relevant to the mechanism of action or site of effect for many therapeutics and immune-mediated diseases.
