Overview

Hantaan virus glycoprotein C (Gc) is a critical structural protein of the Hantaan virus (HTNV), the prototype member of the Orthohantavirus genus and the primary cause of Hemorrhagic Fever with Renal Syndrome (HFRS) (UniProt: P08668; PubMed: 30626688). Gc functions as a class II viral fusion protein, working in a heterodimeric complex with glycoprotein Gn to facilitate viral attachment and entry into host cells (PubMed: 27851987). Following receptor-mediated endocytosis, the low pH environment of the endosome triggers a dramatic conformational change in Gc, leading to the fusion of the viral envelope with the endosomal membrane (PubMed: 31142630). As the major target for neutralizing antibodies, Gc is the focus of vaccine strategies and the development of therapeutic monoclonal antibodies, such as ADI-42898, which aim to block the fusion loop or prevent the transition from a pre-fusion to a post-fusion state (PubMed: 33408178).
