Overview

The HA1 globular head domain of the H5N1 influenza virus hemagglutinin is the primary viral component responsible for recognizing and binding to host cell receptors. It contains the receptor-binding site (RBS), which specifically targets sialic acids on the surface of host respiratory epithelial cells, facilitating viral entry (Skehel & Wiley, 2000). As the most exposed part of the virus, the HA1 head is the principal target for neutralizing antibodies generated by infection or vaccination (Whittle et al., 2011). However, this domain is characterized by high plasticity and a high rate of mutation, a process known as antigenic drift, which allows the virus to evade the host immune system (CDC, 2023). Therapeutic strategies targeting the HA1 head include the development of prepandemic vaccines and monoclonal antibodies designed to block the RBS or stabilize the protein structure to prevent infection. Understanding the structural variations in the H5N1 HA1 domain is crucial for monitoring pandemic potential and ensuring the efficacy of medical countermeasures (UniProt, 2024).
