Overview

The Hemagglutinin protein, H5 subtype is a spike-shaped, homotrimeric glycoprotein on the surface of the influenza A virus, specifically the H5 subtype commonly associated with avian influenza and zoonotic transmission[3][5]. Each HA monomer comprises two polypeptide chains, HA1 and HA2, produced by proteolytic cleavage from a precursor HA0[1][4]. HA mediates viral entry into host cells by binding its receptor-binding domain to sialic acid-containing glycan receptors[3][5], followed by low-pH-triggered fusion of viral and endosomal membranes. The H5 subtype is notable for its ability to cross species barriers after acquiring specific amino acid changes, contributing to pandemic potential[5]. The HA (H5) protein is the primary target for neutralizing antibodies and vaccine development, but its high variability presents challenges for universal vaccine design and sustained efficacy[4][7]. Antibody therapies (including broadly neutralizing monoclonal antibodies) and recombinant vaccine antigens targeting HA (H5) are active areas of influenza drug and vaccine research[2][6][7].
