HLRCC-TC1 (Hereditary Leiomyomatosis and Renal Cell Carcinoma Therapeutic Candidate 1) is a novel cancer-specific therapeutic target identified through integrative genomic profiling of HLRCC-associated renal cell carcinoma. It is specifically overexpressed in HLRCC tumors and other aggressive forms of renal cell carcinoma with poor clinical outcomes, while showing minimal expression in normal tissues. Research indicates that HLRCC-TC1 functions as a driver of cell proliferation in fumarate hydratase (FH)-deficient cells. Experimental targeting of the HLRCC-TC1 mRNA transcript using siRNAs or the resulting protein using neutralizing antibodies has been shown to significantly reduce the growth of HLRCC patient-derived cell lines, such as UOK262. As a tumor-specific marker and vulnerability, it represents a promising candidate for precision medicine in patients with this aggressive and often treatment-resistant form of kidney cancer.
