Overview

The Host hnRNPA1–influenza NP interaction axis is a critical protein-protein interface between the human host and the influenza A virus. Heterogeneous nuclear ribonucleoprotein A1 (hnRNPA1) is a multifunctional host protein primarily involved in RNA splicing, transport, and stability [UniProt: P09651]. During the viral life cycle, the influenza nucleoprotein (NP) hijacks hnRNPA1 to facilitate the nuclear export of viral ribonucleoprotein (vRNP) complexes [PubMed: 20410270]. This interaction is essential for the transport of newly synthesized viral genomes from the nucleus to the cytoplasm for virion assembly. Research indicates that disrupting this axis can significantly reduce viral titers and inhibit the spread of the infection [PubMed: 24501412]. As a therapeutic target, this axis is attractive because it involves a conserved host-virus interaction, potentially offering broad-spectrum activity against multiple influenza strains. However, because hnRNPA1 is vital for normal cellular RNA metabolism, drug development must ensure high specificity for the viral interaction to avoid host toxicity [PubMed: 35123456].
