Overview

High mobility group box 3 (HMGB3) is a non-histone chromosomal protein and a member of the HMGB family that functions as a DNA chaperone, regulating chromatin structure and gene transcription (UniProt P51991). It is essential for the maintenance and self-renewal of hematopoietic stem cells and is highly expressed during embryonic development (NCBI Gene ID: 3149). In various human cancers, such as gastric, breast, and colorectal cancer, HMGB3 mRNA is frequently overexpressed and acts as an oncogene by promoting cell proliferation, migration, and epithelial-mesenchymal transition (PubMed: 28656245, 30106434). Targeting HMGB3 mRNA through RNA interference (RNAi) technologies, including siRNAs and shRNAs, has demonstrated significant potential in preclinical studies to inhibit tumor growth and overcome chemoresistance (PubMed: 25670453). While no FDA-approved drugs currently target HMGB3, it remains a significant candidate for RNA-targeted therapies and serves as a valuable prognostic biomarker in oncology.

Mechanism of action

RNA interference (RNAi) mediated degradation of HMGB3 mRNA, which prevents the translation of the HMGB3 protein and subsequently inhibits oncogenic signaling pathways such as Wnt/beta-catenin and AKT.
