Overview

Histone deacetylases (HDACs) are a family of enzymes that remove acetyl groups from lysine residues on histones and non-histone proteins, leading to chromatin condensation and transcriptional repression. The term "pan-HDAC" refers to agents or inhibitors that target multiple HDAC isoforms across different classes. Inhibition of pan-HDAC results in global hyperacetylation with broad changes in cellular function. They are investigated mainly for oncology indications but also neurological/fibrotic diseases.

Mechanism of action
