Overview

Histone H3.1, encoded by clustered histone genes such as H3C6, is a basic nuclear protein that forms a core component of the nucleosome, the fundamental unit of chromatin structure in eukaryotes. Two molecules each of H2A, H2B, H3 (including H3.1), and H4 form an octamer around which DNA is wrapped into repeating units called nucleosomes, facilitating DNA packaging in the nucleus. Histone H3.1 is replication-dependent and plays central roles in chromosomal stability, regulation of gene expression, DNA replication, and DNA repair through a variety of post-translational modifications such as methylation, acetylation, and phosphorylation. Mutations in histone H3 genes are implicated in cancers, including diffuse midline glioma. Histone H3.1 is not itself a direct therapeutic target (e.g., a "druggable" receptor or enzyme), but its modifications and mutations are key in epigenetic regulation and disease pathogenesis[1][2][3].
