Overview

The HIV-1 envelope glycoprotein gp120-specific B-cell receptor (BCR) is a membrane-bound immunoglobulin complex that serves as the primary recognition unit for the HIV-1 virus on the surface of B lymphocytes. These receptors are the central focus of germline-targeting vaccine strategies, which aim to selectively activate rare B-cell precursors that possess the genetic potential to evolve into broadly neutralizing antibodies (bNAbs) (Jardine et al., Science, 2013). Upon engagement by engineered immunogens like eOD-GT8 60mer, these BCRs initiate intracellular signaling cascades that drive B-cell activation, proliferation, and entry into germinal centers for affinity maturation (Leggat et al., Nature Immunology, 2022). This maturation process is critical for generating antibodies capable of neutralizing a wide array of HIV-1 variants by targeting conserved regions of the gp120 protein, such as the CD4 binding site. Therapeutic development targeting these BCRs is complicated by the high diversity of the human antibody repertoire and the need to avoid inducing autoreactive or polyreactive antibodies (Haynes et al., Nature Biotechnology, 2023). As such, the frequency and specificity of gp120-binding BCRs are used as primary biomarkers to evaluate the success of immunogen design in clinical trials.
