Target intelligence / Profile preview
The HLA-A*02:01-restricted Preferentially expressed Antigen in Melanoma (PRAME) peptide-MHC complex is a specific molecular target formed by the presentation of a PRAME-derived peptide (typically the SLLMWITQC decamer) on the Human Leukocyte Antigen (HLA) allele A*02:01 (PMID: 15958671). PRAME is a well-characterized cancer-testis antigen that is highly expressed in a wide range of malignancies, including melanoma, uveal melanoma, acute myeloid leukemia, and various solid tumors, while its expression in normal tissues is restricted to immune-privileged sites such as the testes (PMID: 28630100). This restricted expression pattern makes the HLA-A*02:01/PRAME complex an ideal target for precision immunotherapy. Current therapeutic strategies include T-cell receptor-engineered T-cell (TCR-T) therapies and bispecific T-cell engagers, such as ImmTACs, which are designed to recognize this specific pMHC complex with high affinity (PMID: 30552139). These drugs, such as IMA203 and IMC-F106C, facilitate the direct killing of cancer cells by redirecting the patient's T-cells to the tumor site. Clinical trials are ongoing to evaluate the efficacy and safety of these agents in patients who are both HLA-A*02:01 positive and have PRAME-expressing tumors.
Targeting of the peptide-MHC complex by engineered T-cell receptors (TCRs) or bispecific TCR-based molecules to induce T-cell mediated lysis of tumor cells.
