The HLA-A*24:02-presented DEP domain-containing protein 1 (DEPDC1) peptide epitope is a tumor-associated antigen complex that serves as a specific target for cancer immunotherapy. DEPDC1 is an oncogene that is significantly overexpressed in various malignancies, including bladder, lung, and esophageal cancers, while showing restricted expression in normal tissues, primarily the testis (Taniuchi et al., 2005; PMID: 16140947). The HLA-A*24:02 allele is one of the most prevalent MHC Class I alleles in East Asian populations, making this specific peptide-HLA complex a strategic target for precision medicine (Obokata et al., 2012; PMID: 22144475). Therapeutic strategies targeting this complex include peptide vaccines, such as DEPDC1-9-294, and T-cell receptor (TCR) engineered T-cell therapies. These treatments aim to stimulate or provide cytotoxic T lymphocytes (CTLs) that recognize the DEPDC1 peptide presented by HLA-A*24:02 on the surface of tumor cells. Upon recognition, the CTLs induce apoptosis in the cancer cells through the release of cytotoxic granules (Yuan et al., 2014; PMID: 24441014). Clinical trials have investigated DEPDC1 peptide vaccines, demonstrating their ability to induce specific immune responses and potentially improve clinical outcomes in patients with DEPDC1-positive tumors (Al-Khadairi et al., 2015; PMID: 26154150). This target represents a promising avenue for personalized immunotherapy in patients who are HLA-A*24:02 positive.
