The HLA-A*24:02-restricted Cell Division Cycle Associated 1 (CDCA1) 56-64 epitope is a specific peptide-major histocompatibility complex (pMHC) target used in cancer immunotherapy. CDCA1, also known as NUF2, is a component of the NDC80 kinetochore complex essential for chromosome segregation during mitosis (UniProt: Q9BZD4). It is classified as a cancer-testis-like antigen because it is highly overexpressed in various malignancies—including lung, bladder, and head and neck cancers—while its expression in normal tissues is restricted primarily to the testis (PubMed: 24333715). The specific epitope (sequence: VYGIRLEHF) is presented by the HLA-A*24:02 allele, which is particularly common in East Asian populations (PubMed: 19536605). Targeting this complex via peptide vaccines or TCR-engineered T-cells (TCR-T) allows the immune system to selectively identify and destroy malignant cells while sparing healthy tissue (PubMed: 22473314). Therapeutic strategies often focus on expanding endogenous CD8+ T-cells that recognize this specific pMHC to elicit a durable anti-tumor response.
