Overview

The HLA-A*24:02-restricted Vascular Endothelial Growth Factor Receptor 1 (VEGFR1) epitope 1084 is a specific peptide-major histocompatibility complex (pMHC) target used in cancer immunotherapy. The epitope is a 9-amino acid sequence (SYGVLLWEI) derived from the VEGFR1 protein, which is presented on the cell surface by the HLA-A*24:02 allele (PMID: 12124337). VEGFR1 is significantly upregulated on the endothelial cells of tumor neovasculature compared to normal vessels, making this complex a selective target for anti-angiogenic therapy (PMID: 15930355). Therapeutic agents, such as peptide vaccines or TCR-engineered T cells, aim to induce cytotoxic T lymphocyte (CTL) responses that specifically recognize and eliminate VEGFR1-expressing endothelial cells (PMID: 21135208). By destroying the tumor's blood supply, this approach inhibits tumor growth and metastasis (PMID: 24583795). This target is particularly relevant for patients carrying the HLA-A*24:02 allele, which is highly prevalent in East Asian populations.

Mechanism of action

Induction of antigen-specific cytotoxic T lymphocytes (CTLs) that recognize the VEGFR1-1084 peptide (SYGVLLWEI) presented by HLA-A*24:02 on tumor-associated endothelial cells, leading to the destruction of tumor vasculature and inhibition of tumor growth (PMID: 12124337, 21135208).
