HLA complex group 11 (HCG11) is a long non-coding RNA (lncRNA) located at chromosome 6p22.2, comprising approximately 5,696 nucleotides and generally transcribed in a single exon[4]. HCG11 is primarily studied for its role as a tumor suppressor in diverse cancer types, including non-small cell lung cancer, hepatocellular carcinoma, prostate cancer, gastric cancer, glioma, and breast cancer. Mechanistically, HCG11 acts via multiple axes, most notably by functioning as a molecular sponge for oncogenic microRNAs such as miR-875, thereby indirectly regulating the expression of tumor suppressor genes like SATB2[1]. Overexpression of HCG11 has been shown to inhibit proliferation, invasion, and migration, while promoting apoptosis and cell cycle arrest in cancer cells[1][2][3]. HCG11 shares regulatory and structural similarity with the lncRNA NORAD, including the presence of Pumilio Recognition Elements (PREs) which interact with RNA-binding proteins, although its direct interaction with therapeutically actionable targets or drugs is currently unreported[4]. The molecule is under active investigation as a prognostic biomarker due to its downregulation in aggressive cancers and its negative correlation with overall survival in certain patient cohorts[1][2][3][4].
