Overview

HLA complex group 18 (HCG18) is a long non-coding RNA gene expressed from the human genome within the HLA region. Unlike classical HLA proteins, HCG18 does not code for an antigen-presenting protein but instead produces a lncRNA transcript. HCG18 acts primarily as a molecular sponge for specific microRNAs (such as miR-324-5p and miR-424-5p), thereby altering the expression of downstream genes (for example, CDK16) that influence cell proliferation, migration, invasion, and apoptosis. Overexpression of HCG18 has been repeatedly observed in tumor tissues such as melanoma and cholangiocarcinoma, where it contributes to aggressive cancer phenotypes and poor survival outcomes. While HCG18 is not a classical drug target, its role in molecular regulation makes it a potential biomarker and a candidate for future RNA-based therapeutic strategies[1][2][3][4].

Mechanism of action

Sponging microRNAs (such as miR-324-5p in melanoma, miR-424-5p in cholangiocarcinoma), resulting in regulation of downstream genes like CDK16. Modulating cell signaling pathways via non-coding RNA interactions.

05

Safety considerations

Not applicable for direct targeting; safety concerns would be theoretical and related to RNA-based therapies or diagnostic use (e.g., off-target effects of RNA therapeutics)
06

Biomarkers

Increased expression of HCG18 in tumors is associated with poor prognosis in several cancers (e.g., melanoma, cholangiocarcinoma)Potential biomarker for cancer diagnosis and prognosis
