Homeobox A10 (HOXA10) mRNA encodes a DNA-binding transcription factor that is a member of the highly conserved homeobox gene family, which plays a fundamental role in regulating gene expression during embryonic development and cellular differentiation (UniProt P31260). In adults, HOXA10 is critically involved in the regulation of uterine receptivity for embryo implantation and the development of the hematopoietic system (Taylor et al., 1998, PMID: 9510025). Dysregulation of HOXA10 mRNA expression is linked to several pathological conditions; for instance, its downregulation in the endometrium is associated with endometriosis and infertility (NCBI Gene ID: 3206). Conversely, its overexpression is a hallmark of certain aggressive leukemias, such as acute myeloid leukemia (AML), where it acts as an oncogene (PubMed: 17360454). As a therapeutic target, HOXA10 mRNA is being explored through the use of RNA-based technologies like antisense oligonucleotides (ASOs) and small interfering RNAs (siRNAs) to modulate its expression in oncogenic contexts (PubChem). However, targeting this molecule presents challenges due to its essential roles in normal physiology and the potential for cross-reactivity with other HOX family members (NIH). Research also suggests that HOXA10 mRNA levels can serve as a biomarker for endometrial receptivity in patients undergoing assisted reproductive technologies (PubMed: 22426357).
