The Homeobox D10 (HOXD10) mRNA 3'-untranslated region (3'-UTR) is a regulatory sequence within the HOXD10 transcript that serves as a primary site for post-transcriptional gene regulation (Ma et al., 2007, Nature). HOXD10 itself is a transcription factor that maintains a differentiated cellular phenotype and suppresses pro-metastatic genes like RHOC (Sun et al., 2011, Brain). In several aggressive cancers, the microRNA miR-10b binds to specific sequences within the HOXD10 3'-UTR, leading to the silencing of HOXD10 expression and subsequent induction of tumor cell invasion and metastasis (Ma et al., 2007, Nature). This interaction makes the HOXD10 3'-UTR a focal point for therapeutic strategies aimed at blocking miR-10b binding using antagomirs or antisense oligonucleotides (NCBI Gene, 3236). By preventing the inhibitory action of miR-10b on this 3'-UTR, these therapeutic agents restore HOXD10 protein levels, thereby inhibiting the metastatic cascade (Sun et al., 2011, Brain). The target is particularly relevant in breast cancer and glioblastoma research, where miR-10b/HOXD10 signaling is a well-characterized driver of disease progression (UniProt, P09071).
