Overview

hsa-miR-182-5p is a member of the miR-183/96/182 cluster located on chromosome 7q32.2 (MDPI, 2023). It is a small non-coding RNA that regulates gene expression post-transcriptionally by binding to the 3'-untranslated regions (UTRs) of target mRNAs, leading to their degradation or translational repression (NIH, 2012). In many cancers, such as breast, colorectal, and hepatocellular carcinoma, it acts as an "oncomir" by targeting tumor suppressor genes like FOXO1 and BRCA1, thereby promoting cell proliferation, survival, and metastasis (NIH, 2023; NIH, 2012). Conversely, it has been shown to act as a tumor suppressor in certain contexts like gastric adenocarcinoma by targeting oncogenes like CREB1 (NIH, 2023). Beyond oncology, hsa-miR-182-5p plays a significant role in the central nervous system, where its upregulation is linked to the pathogenesis of depression through the inhibition of the Akt/GSK3β/CREB signaling pathway and impairment of neurogenesis (NIH, 2023; OUP, 2023). Therapeutic strategies include the use of antagomirs to inhibit its oncogenic activity or mimics to restore its suppressive function, though challenges remain regarding tissue specificity and the modulation of its extensive network of over 1,000 potential gene targets (NIH, 2012).

Mechanism of action

hsa-miR-182-5p functions through post-transcriptional gene silencing by binding to the 3'-untranslated region (3'-UTR) of target mRNAs, leading to mRNA degradation or translational inhibition (NIH, 2012). It modulates key signaling pathways such as PI3K/Akt, Wnt/beta-catenin, and the DNA damage response by targeting genes like BRCA1, FOXO1, and CASP2 (NIH, 2023; NIH, 2019).
