Overview

House dust mite-specific immunoglobulin G4 (HDM-specific IgG4) is a specialized subclass of antibodies produced by B cells in response to prolonged or high-dose exposure to allergens from the dust mite species Dermatophagoides pteronyssinus and Dermatophagoides farinae. In the context of allergic disease, these antibodies serve a protective role by acting as 'blocking antibodies' that compete with allergen-specific IgE for binding to major HDM allergens such as Der p 1, Der p 2, and Der f 1 (PMID: 30103933). By intercepting these allergens, IgG4 prevents the cross-linking of IgE bound to mast cells and basophils, which otherwise triggers the release of inflammatory mediators like histamine and leukotrienes (PMID: 28212950). Beyond simple neutralization, HDM-specific IgG4 also inhibits IgE-facilitated antigen presentation to T cells, thereby dampening the overall Th2-mediated allergic cascade (PMID: 21834850). The induction of high levels of HDM-specific IgG4 is a primary goal and a key biomarker of clinical efficacy in allergen immunotherapy (AIT), signaling a shift toward immunological tolerance. Therapeutic products such as Acarizax and Odactra are specifically designed to stimulate the production of these antibodies to provide long-term relief from allergic rhinitis and asthma symptoms.
