Overview

Human chorionic gonadotropin subunit beta (hCG-beta) is a glycoprotein hormone produced primarily by syncytiotrophoblast cells in the placenta during pregnancy (UniProt P01233). It is the unique subunit of the hCG heterodimer, providing biological specificity compared to the alpha subunit shared with LH, FSH, and TSH (StatPearls, 2023). Its primary physiological role is to maintain the corpus luteum, ensuring continued progesterone production essential for early pregnancy maintenance (PubMed, PMID: 25233380). Beyond pregnancy, hCG-beta is frequently overexpressed in various malignancies, including germ cell tumors and certain epithelial cancers, where it may promote tumor cell survival and angiogenesis (PubMed, PMID: 21709442). Therapeutically, recombinant or urinary-derived hCG is used to induce ovulation in infertility treatments and to treat hypogonadism in males by stimulating testosterone production (DrugBank DB00097). As a clinical biomarker, it is the gold standard for pregnancy testing and a critical tool for monitoring gestational trophoblastic diseases and certain cancers (StatPearls, 2023).

Mechanism of action

hCG-beta, as part of the hCG heterodimer, binds to and activates the Luteinizing Hormone/Chorionic Gonadotropin Receptor (LHCGR), a G protein-coupled receptor (DrugBank DB00097). This interaction mimics the action of luteinizing hormone (LH), stimulating the corpus luteum to produce progesterone in females and Leydig cells to produce testosterone in males (StatPearls, 2023).
