Overview

The HLA-A2–presented CS1(239-247) epitope is a specific peptide-major histocompatibility complex (pMHC) target used in the immunotherapy of multiple myeloma (Bae et al., 2012). CS1, also known as SLAMF7 or CD319, is a cell surface glycoprotein highly and nearly universally expressed on malignant plasma cells (UniProt Q9NQ25). The specific nonamer peptide sequence (SLFVLGLFL) derived from the CS1 protein is processed and presented by the HLA-A*02:01 molecule, making it a visible target for the cellular immune system. Therapeutic strategies targeting this epitope include the development of T-cell receptor (TCR) engineered T cells and peptide-based vaccines designed to elicit a robust cytotoxic T lymphocyte (CTL) response. Because CS1 is also expressed at lower levels on some normal immune cells like Natural Killer (NK) cells and certain T-cell subsets, safety monitoring for hematopoietic fratricide or lymphopenia is a critical consideration. This target represents a precision medicine approach, requiring patients to be both HLA-A*02:01 positive and have CS1-expressing tumors.
