Overview

The Human Leukocyte Antigen A*24:02-restricted Cell Division Cycle Associated 1 peptide complex is a specific antigen-presenting structure formed by the HLA-A*24:02 allele and a peptide fragment (typically CDCA1-65, VYGFVRACL) derived from the Cell Division Cycle Associated 1 (CDCA1) protein, also known as NUF2 (UniProt: O14777). CDCA1 is a critical component of the Ndc80 kinetochore complex, essential for chromosome segregation during mitosis (PubMed: 16849515). This protein is highly overexpressed in various cancers, such as non-small cell lung cancer and gastric cancer, while its expression in normal tissues is restricted primarily to the testis, making the pMHC complex a highly specific target for immunotherapy (PubMed: 21860410). The HLA-A*24:02 allele is particularly prevalent in East Asian populations, which enhances the clinical relevance of this target in those regions (Allele Frequency Net Database). Therapeutic strategies targeting this complex include peptide vaccines designed to elicit endogenous cytotoxic T-cell responses and engineered T-cell receptor (TCR-T) therapies that provide direct tumor recognition (ClinicalTrials.gov: NCT01223235).
