The HLA-A*24:02-TTK-567 peptide complex is a specific molecular target used in cancer immunotherapy, particularly for patients carrying the HLA-A*24:02 allele, which is prevalent in Asian populations. The complex consists of the HLA-A*24:02 molecule presenting a 9-amino acid epitope (SYVNEHTYM) derived from the Threonine and Tyrosine Kinase (TTK), also known as Monopolar Spindle 1 (Mps1). TTK is a critical component of the spindle assembly checkpoint and is frequently overexpressed in various malignant tumors while remaining nearly absent in normal adult tissues, making it an ideal tumor-associated antigen. When this peptide is presented in the HLA-A*24:02 binding groove, it is recognized by the T-cell receptors of CD8+ cytotoxic T lymphocytes, triggering an immune-mediated attack against the cancer cells. Therapeutic approaches targeting this complex include peptide-based vaccines and TCR-engineered T-cell therapies. Clinical studies have explored these interventions in several solid tumors, demonstrating the ability to induce specific immune responses with a favorable safety profile.
The complex acts as a ligand for specific T-cell receptors (TCRs) on cytotoxic T lymphocytes (CTLs). Therapeutic strategies involve using the TTK-567 peptide as a vaccine to induce the expansion of CTLs that specifically recognize and kill tumor cells presenting this epitope in the HLA-A*24:02 groove. Alternatively, engineered TCR-T cells can be designed to bind directly to this peptide-MHC complex to initiate tumor lysis.
