Overview

The Human papillomavirus (HPV) L1 capsid protein is the primary structural component of the HPV virion, forming the outer shell that protects the viral genome (UniProt: P03101). It self-assembles into virus-like particles (VLPs) that are highly immunogenic but non-infectious, serving as the basis for prophylactic vaccines (CDC, 2021). Biologically, L1 mediates the initial binding of the virus to heparan sulfate proteoglycans on the host cell surface, which is essential for viral entry and infection (PubMed: 25693011). The specific types 6, 11, 16, 18, 31, 33, 45, 52, and 58 are targeted because they represent the most oncogenic and prevalent strains; high-risk types (16, 18, 31, 33, 45, 52, 58) cause the majority of cervical cancers, while low-risk types (6, 11) cause approximately 90% of genital warts (NCI, 2023). Vaccines like Gardasil 9 target these L1 proteins to induce neutralizing antibodies that block the virus before it can infect the basal layer of the epithelium. This mechanism provides long-term protection against HPV-related malignancies and benign lesions (FDA, 2020).
