The Human papillomavirus type 18 (HPV18) L1 virus-like particle (VLP) is a recombinant protein assembly that mimics the outer shell of the HPV18 virus without containing infectious genetic material (NCBI, 2023). HPV18 is a high-risk genotype responsible for a significant portion of cervical, anogenital, and oropharyngeal cancers worldwide (WHO, 2022). The L1 protein is the major capsid protein of the virus and has the intrinsic ability to self-assemble into VLPs that are antigenically identical to the native virion (UniProt P06794). These VLPs serve as the primary immunogenic component in prophylactic vaccines such as Gardasil 9 and Cervarix, which aim to prevent infection by inducing a robust humoral immune response (FDA, 2020). Upon administration, the immune system generates neutralizing antibodies that target the L1 protein, effectively blocking the virus from attaching to and entering host epithelial cells during subsequent exposure (PubMed, PMID: 28935227). This prevention of persistent infection is a critical strategy in reducing the global burden of HPV-associated malignancies. Because these particles lack viral DNA, they are non-infectious and non-oncogenic, providing a safe and effective method for long-term immunization (StatPearls, 2023).
The mechanism of action involves active immunization where the VLP acts as an antigen to stimulate the production of type-specific neutralizing antibodies; these antibodies bind to the L1 protein on the surface of the HPV18 virion, preventing viral attachment to the basement membrane and subsequent entry into basal keratinocytes (StatPearls, 2023; PubMed, PMID: 28935227).
