Overview

Hyaluronan, also known as hyaluronic acid, is a high-molecular-weight glycosaminoglycan (acid mucopolysaccharide) that serves as a fundamental structural component of the extracellular matrix (ECM) in vertebrates (StatPearls: Hyaluronic Acid, 2023). It is unique among glycosaminoglycans because it is non-sulfated and synthesized by hyaluronan synthases at the inner surface of the plasma membrane rather than being processed in the Golgi apparatus (PubChem: Hyaluronic Acid, CID 24759). Biologically, it regulates tissue hydrodynamics, provides mechanical cushioning in joints, and modulates cell behavior through interactions with receptors like CD44 and RHAMM (NCBI: Glycosaminoglycans, 2022). In pathology, hyaluronan is implicated in cancer metastasis, chronic inflammation, and the progression of osteoarthritis (Journal of Biological Chemistry: Hyaluronan functions, 2016). Therapeutic strategies involve either the administration of exogenous hyaluronan for viscosupplementation and wound healing or the use of hyaluronidase enzymes to degrade the ECM barrier for improved drug delivery (StatPearls: Hyaluronic Acid, 2023). Additionally, hyaluronan levels in the serum serve as important biomarkers for liver fibrosis and other inflammatory conditions (NCBI: Hyaluronan as a Biomarker, 2021).
