Overview

Other hyaluronan receptors refers to a group of cell surface and intracellular proteins that bind hyaluronan (HA) but are distinct from the primary receptor CD44. This group includes the Receptor for Hyaluronan-Mediated Motility (RHAMM/HMMR), Lymphatic Vessel Endothelial Hyaluronan Receptor 1 (LYVE-1), Hyaluronan Receptor for Endocytosis (HARE/Stabilin-2), and Layilin (LAYN). These receptors are involved in a wide range of physiological processes, including the systemic clearance of HA from the blood and lymph (HARE), the trafficking of immune cells through lymphatic vessels (LYVE-1), and the regulation of cell migration, proliferation, and intracellular signaling (RHAMM and Layilin). In pathological conditions such as cancer, these receptors are often overexpressed and contribute to tumor progression, metastasis, and resistance to therapy. Therapeutic targeting of these receptors is an active area of research, with approaches including peptide vaccines (e.g., RHAMM-R3), monoclonal antibodies, and blocking peptides designed to disrupt HA-mediated signaling or modulate HA turnover.

Blocking of hyaluronan-receptor interactions to inhibit downstream signaling pathways such as ERK1/2 and PI3K/AKT, or inhibiting the systemic clearance of hyaluronan to modulate its biological effects and prevent tumor cell lodging.
