The hypothalamus is a small but critical region of the brain that acts as the central command center for thermoregulation, maintaining core body temperature within a narrow, optimal range. It integrates signals from both internal and external sources to coordinate physiological responses that balance heat production and loss. Key areas involved are the Preoptic Area (POA) and Dorsomedial Hypothalamus (DMH). The POA contains warm-sensitive neurons that initiate cooling responses, while the DMH contains cold-sensitive neurons and promotes heat production. TRP channels like TRPM8 (cold sensor) and TRPV1/V3/V4/TRPM2 (potential warm sensors) are involved in sensing temperature changes. Failure of hypothalamic thermoregulation can result in life-threatening conditions such as hyperthermia or hypothermia.
