Ikaros family zinc finger protein 4 (IKZF4), also known as Eos, is a transcription factor that plays a pivotal role in the maintenance of regulatory T cell (Treg) identity and suppressive function (UniProt Q9H2S9). It functions by forming complexes with Foxp3 and C-terminal binding protein (CtBP) to repress the expression of effector cytokines like IL-2 and IFN-gamma, thereby preventing Tregs from acquiring a pro-inflammatory phenotype (Pan et al., 2009, Science). In the context of cancer, IKZF4 is a target for immunotherapy because its genetic or pharmacological silencing can reprogram intratumoral Tregs into effector-like cells, enhancing the anti-tumor immune response (Sharma et al., 2013, Immunity). Additionally, IKZF4 has been implicated in the pathogenesis of autoimmune diseases, such as systemic lupus erythematosus (SLE), where its downregulation is associated with defective Treg function (NCBI Gene ID: 64131). Therapeutic strategies targeting IKZF4 mRNA, such as siRNA or antisense oligonucleotides, are currently being explored to modulate immune responses in both oncology and inflammatory conditions.
