Immunoglobulin E (IgE) specific for Olea europaea allergens represents a subset of antibodies that mediate Type I hypersensitivity reactions to olive tree pollen, a major cause of respiratory allergy in Mediterranean climates (Bousquet et al., 2001). These antibodies bind to specific proteins from the olive tree, most notably the major allergen Ole e 1, as well as others like Ole e 2 (profilin) and Ole e 9 (WHO/IUIS Allergen Nomenclature). Upon exposure to olive pollen, these IgE molecules cross-link with high-affinity FcεRI receptors on the surface of mast cells and basophils, triggering the release of inflammatory mediators such as histamine and leukotrienes. This process leads to clinical symptoms of allergic rhinitis, conjunctivitis, and bronchial asthma. Therapeutic strategies targeting this pathway include the use of monoclonal antibodies like omalizumab, which sequesters free IgE to prevent receptor binding, and allergen-specific immunotherapy (AIT), which aims to desensitize the immune system through controlled exposure to Olea europaea extracts (Valenta et al., 2016). Monitoring serum levels of these specific IgE antibodies is a standard diagnostic practice for identifying sensitized individuals and managing treatment progress.
Omalizumab binds to the Cε3 domain of IgE, preventing its interaction with the high-affinity IgE receptor (FcεRI) on mast cells and basophils (StatPearls, 2023). Allergen-specific immunotherapy (AIT) involves the administration of increasing doses of Olea europaea allergens to induce immunological tolerance, shifting the immune response from a Th2-mediated allergic profile to a Th1/Treg-mediated protective profile, often characterized by the production of IgG4 blocking antibodies (Valenta et al., 2016).
