Prawn tropomyosin-specific IgE refers to the subset of Immunoglobulin E antibodies that recognize and bind to tropomyosin, the major heat-stable allergen found in prawns and other crustaceans, such as Pen m 1 from Penaeus monodon (PMID: 8952561). In sensitized individuals, these antibodies are primarily bound to high-affinity FcεRI receptors on the surface of mast cells and basophils (PMID: 18294912). When the individual ingests prawn, the tropomyosin allergen cross-links these surface-bound IgE molecules, triggering the immediate release of potent inflammatory mediators like histamine, prostaglandins, and leukotrienes (PMID: 11204551). This process is the underlying mechanism of Type I hypersensitivity reactions, which can manifest as oral allergy syndrome, gastrointestinal distress, or systemic anaphylaxis (PMID: 10424233). While broad-spectrum anti-IgE therapies like Omalizumab can reduce the overall IgE pool by binding the Cε3 domain, specific IgE levels are primarily used as diagnostic biomarkers to assess the severity and likelihood of clinical reactions (PMID: 24414352). Emerging treatments also focus on allergen-specific immunotherapy (ASIT) to shift the immune response from IgE production toward protective IgG4 antibodies (PMID: 21235890).
Monoclonal antibodies like Omalizumab bind to the Cε3 domain of circulating IgE, preventing its interaction with the high-affinity IgE receptor (FcεRI) on mast cells and basophils, thereby preventing degranulation (PMID: 18294912). Additionally, allergen-specific immunotherapy aims to desensitize the patient by inducing regulatory T cells and IgG4 antibodies that compete with IgE for allergen binding (PMID: 21235890).
