Overview

Immunoglobulin E (IgE) antibodies specific to peanut allergens Ara h 1, Ara h 2, and Ara h 3 are produced by the immune system of individuals sensitized to peanuts. These antibodies bind to particular epitopes (regions) on the allergen proteins and, upon re-exposure to peanut, cross-link IgE receptors (FcεRI) on mast cells and basophils, triggering the release of histamine and other mediators responsible for immediate hypersensitivity reactions, including anaphylaxis[1][2][3][5][6]. Circulating levels of IgE against Ara h 2 in particular are correlated with the clinical severity of peanut allergy, and these antibodies serve as key biomarkers in the diagnosis, prognosis, and monitoring of peanut allergy. Therapeutic targeting of these IgE molecules (for example, with anti-IgE monoclonal antibodies such as omalizumab) can decrease allergic reactivity and is under investigation for allergen immunotherapy. The specificity for individual allergen components (Ara h 1, h 2, h 3) determines the individual sensitization profile and may guide both treatment approaches and risk assessment[1][2][4][5][6].
