Overview

Immunoglobulin heavy variable 3-16 (IGHV3-16) is a gene segment of the immunoglobulin heavy chain variable region locus that encodes a probable non-functional open reading frame (ORF) of the variable (V) region of immunoglobulin heavy chains[3][6]. While the IGHV gene family is essential for antibody diversity in the adaptive immune system, IGHV3-16 is classified as a pseudogene or non-functional segment. Unlike functional IGHV genes, it does not produce a productive immunoglobulin chain due to alterations in V-(D)-J recombination sites, splicing errors at the mRNA level, or conserved amino acid changes at the protein level[3]. Immunoglobulins (antibodies) are critical for humoral immunity, but this non-functional IGHV segment cannot directly participate in immune signaling or serve as a therapeutic target. The IGHV locus as a whole is highly polymorphic and relevant for immune repertoire diversity, but IGHV3-16 itself does not code for a biologically active protein involved in disease or targeted therapy[2][4][5][3][6].
