IMX313 is a protein multimerization domain derived from the C-terminal region of the chicken C4b-binding protein (C4bp) [PMID: 18337592, PMID: 22438547]. It is engineered to self-assemble into stable heptameric structures, which allows for the multivalent display of antigens when genetically fused to the domain [PMID: 26743453, PMID: 34305935]. This repetitive arrangement mimics the structural patterns found on the surfaces of many viruses and bacteria, thereby facilitating the cross-linking of B-cell receptors and significantly enhancing the activation of the adaptive immune system [PMID: 26743453]. IMX313 is primarily utilized as a vaccine platform technology to increase the potency and breadth of immune responses against various infectious diseases, including malaria, tuberculosis, and influenza [PMID: 22438547, PMID: 35655174]. By improving antigen stability and promoting more efficient uptake by antigen-presenting cells, it serves as a molecular adjuvant that can elicit robust antibody and T-cell responses even at low antigen doses [PMID: 18337592, PMID: 22438547]. Clinical trials have demonstrated that IMX313-based vaccines are generally safe and do not induce cross-reactive antibodies against human C4bp due to low sequence homology [PMID: 34305935].
