Overview

Influenza A virus neuraminidase (subtype N1) is a critical surface glycoprotein and enzyme found on the envelope of influenza A viruses. Its primary biological function is the cleavage of terminal sialic acid residues from host cell receptors and viral glycoproteins, which is essential for the release of newly formed virions from the surface of infected cells (UniProt, 2023). By preventing the aggregation of progeny viruses and facilitating their movement through the respiratory mucus, N1 neuraminidase enables the efficient spread of the infection throughout the host (PubMed, PMC3342934). In the context of disease, N1 is a defining component of highly pathogenic and pandemic strains, such as the H1N1 and H5N1 subtypes (CDC, 2022). It serves as a major target for antiviral drugs known as neuraminidase inhibitors, including oseltamivir and zanamivir, which bind to the enzyme's conserved active site to block its catalytic activity (StatPearls, 2023). The clinical utility of these drugs is frequently challenged by the emergence of resistance-conferring mutations, most notably the H275Y substitution, which significantly reduces drug binding affinity (Nature, 2012).

Competitive inhibition of the neuraminidase enzyme active site, which prevents the cleavage of terminal sialic acid residues on host cell receptors, thereby trapping progeny virions on the cell surface and halting viral spread.
