Overview

The influenza virus hemagglutinin (HA) head domain is the distal, globular portion of the HA surface glycoprotein, primarily composed of the HA1 subunit (UniProt P03435). It contains the receptor-binding site (RBS) that facilitates viral attachment to sialic acid residues on the surface of host respiratory epithelial cells (PubMed: 22226355). In the context of seasonal influenza vaccines, the HA head domain is the principal antigen used to elicit neutralizing antibodies (CDC, 2023). These antibodies typically function by sterically hindering the RBS, thereby preventing the virus from initiating the infection cycle (Nature Reviews Microbiology, 2019). However, the head domain is highly susceptible to antigenic drift, where point mutations in the epitopes allow the virus to evade pre-existing immunity, necessitating the annual reformulation of vaccines to include current circulating strains (WHO, 2024). While highly effective at inducing a potent immune response, the focus on the head domain often results in narrow, strain-specific protection rather than broad-spectrum immunity against diverse influenza subtypes (PubMed: 30104474). Vaccine-included strains typically represent the most prevalent H1N1, H3N2, and Influenza B lineages identified through global surveillance (CDC, 2024). Therapeutic strategies targeting this domain include both inactivated and recombinant vaccines, as well as monoclonal antibodies designed to provide immediate passive immunity (PubMed: 29439166).
