Overview

The innate immune pattern recognition pathways activated by the NE01 nanoemulsion adjuvant (also known as W805EC) represent a critical mechanism for enhancing vaccine immunogenicity, particularly in mucosal settings. NE01 is a surfactant-stabilized, oil-in-water nanoemulsion that, upon intranasal administration, induces localized cellular stress and immunogenic cell death (ICD) in the nasal epithelium (Lindell et al., 2011, Journal of Immunology). This process leads to the release of endogenous danger-associated molecular patterns (DAMPs), which are subsequently sensed by pattern recognition receptors (PRRs) such as the NLRP3 inflammasome and potentially Toll-like receptors like TLR2 and TLR4 (Bielinska et al., 2014, Hum Vaccin Immunother). Activation of these pathways triggers the maturation of pro-inflammatory cytokines, including IL-1β and IL-18, and facilitates the recruitment and activation of dendritic cells. Consequently, NE01 promotes a robust and balanced immune response involving Th1 and Th17 cellular immunity, as well as high titers of systemic IgG and mucosal IgA antibodies (Makidon et al., 2008, PLoS ONE). This target pathway is primarily utilized in the development of vaccines against respiratory pathogens like influenza, respiratory syncytial virus (RSV), and SARS-CoV-2.

Mechanism of action

NE01 activates the NLRP3 inflammasome and other pattern recognition receptors by inducing local cellular stress and the release of damage-associated molecular patterns (DAMPs), which enhances antigen presentation and promotes Th1/Th17/IgA immune responses.
