Inositol 1,4,5-trisphosphate receptor-interacting protein (ITPRIP) is a membrane-associated protein that binds to and modulates the activity of the inositol 1,4,5-trisphosphate receptor (ITPR), a key channel involved in calcium signaling. ITPRIP enhances the sensitivity of ITPR to intracellular Ca²⁺ and allosterically modulates its inhibition by calcium, which in turn affects various calcium-dependent cellular processes. It is expressed prominently in neuronal and mesenchymal cells, localizing to peripheral membranes and the outer nuclear membrane. ITPRIP participates in diverse biological contexts, including regulating cytosolic calcium balance, modulating the innate antiviral response, influencing cell cycle and cell death pathways, supporting viral vector replication efficiency, and contributing to metabolic adaptation under hypoxic stress. Altered ITPRIP expression and function have been implicated in cancers, neurodevelopmental disorders, neurodegenerative diseases, and responses to viral infection[1][2][3][4].
