Overview

Inositol 1,4,5-trisphosphate receptor-interacting protein-like 1 (ITPRIPL1) functions as a ligand of the CD3E subunit of the T cell receptor complex, inhibiting TCR-CD3 signaling and thereby regulating T cell activation. It induces stable binding between CD3E and NCK1, preventing the association of CD3E with other cytoplasmic partners following T cell activation. While the protein interacts with the IP3 receptor complex, it acts primarily as a regulatory/adaptor protein rather than as a channel, enzyme, or classical receptor. This protein should not be confused with the inositol 1,4,5-trisphosphate receptor itself (IP3R or ITPR), which is an intracellular Ca²⁺ channel critical for calcium signaling pathways; ITPRIPL1's name refers to its interacting and regulatory function, not the receptor channel. ITPRIPL1 is also distinct from 'Inositol 1,4,5-trisphosphate receptor-interacting protein' (ITPRIP), a separate but related regulatory protein. There is currently no evidence linking ITPRIPL1 to specific drugs, biomarker use, or direct roles in therapeutic safety or disease development, although its participation in immune signaling pathways (T cell regulation) suggests relevance to immunology research.
