Interferon-gamma (IFN-γ)-producing T cells, primarily comprising Th1 CD4+ cells and CD8+ cytotoxic T lymphocytes, are central coordinators of the adaptive immune response against intracellular pathogens and tumors (PMID: 14734551). These cells secrete IFN-γ, a potent pleiotropic cytokine that activates macrophages, enhances antigen presentation via MHC class I and II induction, and promotes the differentiation of more Th1 cells (PMID: 29780381). Conversely, dysregulated or chronic activation of IFN-γ-producing T cells is a hallmark of various autoimmune and inflammatory conditions, such as rheumatoid arthritis and multiple sclerosis (PMID: 30217907). Therapeutic strategies involve either boosting their activity to combat cancer and infections or suppressing them to treat inflammatory disorders. Monitoring these cells via biomarkers like T-bet expression or IFN-γ secretion levels is critical for assessing treatment efficacy in modern immunotherapy.
Therapeutic modulation involves the activation and expansion of these cells to enhance antitumor immunity (e.g., via checkpoint inhibition) or their suppression to control inflammatory and autoimmune responses.
