Interferon-induced protein with tetratricopeptide repeats 3 (IFIT3) mRNA is a transcript of the IFIT3 gene, which belongs to a family of interferon-stimulated genes (ISGs) critical for the host's innate antiviral response (UniProt P15812). The protein encoded by this mRNA acts as a molecular scaffold, bridging signaling molecules like MAVS and RIG-I to amplify the production of type I interferons (Liu et al., 2011, PMID: 21478870). Elevated levels of IFIT3 mRNA are a hallmark of the "interferon signature" observed in autoimmune conditions such as systemic lupus erythematosus (SLE) and Sjögren's syndrome (Rose et al., 2013, PMID: 23454755). In the context of oncology, IFIT3 mRNA overexpression has been linked to increased cell proliferation, metastasis, and resistance to chemotherapy in various cancers, including pancreatic and breast cancer (Yang et al., 2018, PMID: 29654258). While there are currently no FDA-approved drugs that directly target IFIT3 mRNA, it is an active area of research for RNA-based therapeutics like siRNAs and antisense oligonucleotides. Modulating IFIT3 mRNA levels offers a potential therapeutic avenue for treating both chronic inflammatory diseases and certain aggressive malignancies. Interferon-based therapies, such as Peginterferon alfa-2a, are known to potently induce IFIT3 mRNA expression as part of their therapeutic mechanism (NIH, PubChem). Understanding the regulation of IFIT3 mRNA is essential for developing strategies to treat chronic inflammation and viral evasion.
