The Interferon regulatory factor 8 (IRF8) messenger RNA 3' untranslated region (3'UTR) is a critical regulatory segment of the IRF8 transcript, which encodes a transcription factor essential for the development of myeloid and lymphoid lineages (UniProt P62817). This region contains multiple binding sites for microRNAs, such as miR-22 and miR-146a, and RNA-binding proteins that post-transcriptionally control IRF8 protein levels by influencing mRNA stability and translation (PubMed: 20810918, PubMed: 26437316). In oncology, particularly in myeloid leukemias, the IRF8 3'UTR is often targeted by overexpressed microRNAs that silence IRF8, thereby acting as a tumor suppressor whose loss facilitates leukemic progression (PubMed: 17438064). Conversely, genetic variations or dysregulated processing of this UTR are implicated in autoimmune susceptibility, including Multiple Sclerosis and Systemic Lupus Erythematosus (PubMed: 21833088). Therapeutic strategies targeting this region involve the use of antisense oligonucleotides (ASOs) or microRNA modulators to either restore IRF8 expression in cancer or dampen it in inflammatory contexts. As a therapeutic target, it offers a precise mechanism to tune the expression of a master immune regulator without direct genomic modification. Research into small molecules that can bind specific RNA secondary structures within the 3'UTR is also an emerging area of drug discovery (PubMed: 32355861).
